What is KPV?
KPV is a tripeptide, just three amino acids: lysine, proline, and valine. It is the tail end (residues 11 to 13) of alpha-MSH, a hormone your body uses to control both inflammation and skin pigmentation. Researchers isolated this fragment because it keeps the anti-inflammatory punch of the parent hormone without the skin-darkening effect. It is a research compound, studied mostly in cell cultures and animal models of gut and skin inflammation, and no approved human product uses it.
How KPV works
KPV calms inflammation from inside the cell. It blocks the nuclear translocation of NF-kB, the master switch that turns on inflammatory cytokine genes, and it also dampens MAP-kinase signaling. Both effects show up at low, nanomolar concentrations in lab studies. There is a useful quirk to its size: because it is only three amino acids, KPV can hitch a ride on the PepT1 transporter that intestinal cells use to absorb small peptides. In mouse models of colitis, KPV reduced inflammation and tissue damage, and it worked in immune cells that do not carry the classic melanocortin receptors, which points to an intracellular route rather than the usual alpha-MSH pathway. The honest gap is that there are no published human clinical trials, so the promising animal and cell data has never been tested in people at any real scale.
