Semaglutide Reconstitution
How to mix Semaglutide and read the exact units to draw. Enter your vial size and bacteriostatic water below for the concentration and dose on a U-100 insulin syringe.
How to reconstitute Semaglutide
- Swab the rubber stopper of the Semaglutide vial and the bacteriostatic water vial with an alcohol wipe.
- Draw 2 ml of bacteriostatic water into a syringe.
- Inject it slowly down the inside wall of the Semaglutide vial, not directly onto the powder.
- Swirl gently until the powder fully dissolves. Do not shake. Shaking can damage the peptide.
- No manufacturer stability data exists for reconstituted lyophilised semaglutide, because no approved product is supplied that way.
Semaglutide concentration by water volume
Semaglutide is commonly supplied in 5 mg vials. The concentration depends only on how much bacteriostatic water you add. More water lowers the concentration and makes small doses easier to measure.
| BAC water added | Concentration | Per 1 unit |
|---|---|---|
| 1 ml | 5,000 mcg/ml | 50 mcg |
| 2 ml | 2,500 mcg/ml | 25 mcg |
| 3 ml | 1,666.7 mcg/ml | 16.7 mcg |
Semaglutide units to draw
At 2,500 mcg/ml (a 5 mg vial in 2 ml of bacteriostatic water), here is the draw for doses across the range Semaglutide is typically used at. Change any number in the calculator above for your own vial.
| Dose | Units to draw | Volume |
|---|---|---|
| 250 mcg | 10 units | 0.10 ml |
| 500 mcg | 20 units | 0.20 ml |
| 1 mg | 40 units | 0.40 ml |
| 1.7 mg | 68 units | 0.68 ml |
| 2.4 mg | 96 units | 0.96 ml |
Semaglutide dosing
Semaglutide follows a fixed, gradual titration. The standard weight-management schedule steps up every four weeks: 0.25 mg, then 0.5, 1.0, 1.7, and finally 2.4 mg weekly, reaching the full dose around week 16 to 20. The 0.25 mg starting dose is not therapeutic. It exists purely to let your gut adapt and blunt the nausea. If a step is rough, the guidance is to hold or slow down rather than push through, because rushing the titration does not speed up results. Enter your own vial size, water volume, and target dose in the calculator above to convert any Semaglutide protocol into exact insulin-syringe units. Storing reconstituted Semaglutide
No manufacturer stability data exists for reconstituted lyophilised semaglutide, because no approved product is supplied that way. Refrigerate at 2 to 8 degrees C and protect from light. For reference, the Ozempic pen may be kept 56 days after first use, but that is a different physical product and the figure does not transfer.
Approved semaglutide ships as a prefilled pen or a ready-to-use solution vial, never as a powder you reconstitute. Lyophilised vials come from the research-supply and compounding channel, so the vial size and water volume here are convention, not label values. Approved titration is 0.25 mg weekly for 4 weeks, then 0.5, 1.0, 1.7, and 2.4 mg at 4-week intervals.
The dose range above comes from approved prescribing information for Semaglutide.
The math
- Concentration (mcg/ml) = vial size in mg × 1,000 ÷ bacteriostatic water in ml.
- Volume for your dose (ml) = dose in mcg ÷ concentration.
- Units to draw = volume in ml × 100 on a U-100 insulin syringe.

Semaglutide reconstitution FAQ
Draw 2 ml of bacteriostatic water into a syringe, inject it slowly down the inside wall of the Semaglutide vial, and swirl gently until the powder dissolves. Do not shake. The concentration is the vial size in mg times 1,000, divided by the millilitres you added.
2 ml for a 5 mg vial gives 2,500 mcg/ml. More liquid makes small doses easier to measure accurately. The calculator shows the concentration for any volume you choose.
Units to draw equals your dose in mcg divided by the concentration in mcg/ml, times 100 on a U-100 insulin syringe. At 2,500 mcg/ml, a 1 mg dose is 40 units.
